The London Legacy Development Corporation (LLDC) has received the green light for the construction of five new neighbourhoods across 64 hectares (158 acres) of the Olympic Park.
Community spaces - including gyms and libraries – will form part of the new developments, which have been approved by the Olympic Delivery Authority's planning committee.
The decision means that LLDC remains on course to establish the first new neighbourhood at Chobham Manor following the 2012 Games, which is due for completion by late 2014.
LLDC chief executive Andrew Altman said: "We can now realise the vision of creating a new piece of the city that will transform people's lives with new opportunities."
London mayor Boris Johnson added: "This is a huge step forward in turning the vision for the development of Queen Elizabeth Olympic Park into a reality."
